    Mitochondrial Dynamics Controls T Cell Fate Through Metabolic Programming

    Activated effector T (TE) cells augment anabolic pathways of metabolism, such as aerobic glycolysis, while memory T (TM) cells engage catabolic pathways, like fatty acid oxidation (FAO). However, signals that drive these differences remain unclear. Mitochondria are metabolic organelles that actively transform their ultrastructure. Therefore, we questioned whether mitochondrial dynamics controls T cell metabolism. We show that TE cells have punctate mitochondria, while TM cells maintain fused networks. The fusion protein Opa1 is required for TM, but not TE cells after infection, and enforcing fusion in TE cells imposes TM cell characteristics and enhances antitumor function. Our data suggest that, by altering cristae morphology, fusion in TM cells configures electron transport chain (ETC) complex associations favoring oxidative phosphorylation (OXPHOS) and FAO, while fission in TE cells leads to cristae expansion, reducing ETC efficiency and promoting aerobic glycolysis. Thus mitochondrial remodeling is a signaling mechanism that instructs T cell metabolic programming

    Reconstructions based directly upon forensic evidence alone are called primary information. Historically this consists of documentation of findings by verbal protocols, photographs and other visual means. Currently modern imaging techniques such as 3D surface scanning and radiological methods (Computer Tomography, Magnetic Resonance Imaging) are also applied. Secondary interpretation is based on facts and the examiner's experience. Usually such reconstructive expertises are given in written form, and are often enhanced by sketches. However, narrative interpretations can, especially in complex courses of action, be difficult to present and can be misunderstood. In this report we demonstrate the use of graphic reconstruction of secondary interpretation with supporting pictorial evidence, applying digital visualisation (using ‘Poser') or scientific animation (using ‘3D Studio Max', ‘Maya') and present methods of clearly distinguishing between factual documentation and examiners' interpretation based on three cases. The first case involved a pedestrian who was initially struck by a car on a motorway and was then run over by a second car. The second case involved a suicidal gunshot to the head with a rifle, in which the trigger was pushed with a rod. The third case dealt with a collision between two motorcycles. Pictorial reconstruction of the secondary interpretation of these cases has several advantages. The images enable an immediate overview, give rise to enhanced clarity, and compel the examiner to look at all details if he or she is to create a complete imag

    ArchAlign: coordinate-free chromatin alignment reveals novel architectures

    To facilitate identification and characterization of genomic functional elements, we have developed a chromatin architecture alignment algorithm (ArchAlign). ArchAlign identifies shared chromatin structural patterns from high-resolution chromatin structural datasets derived from next-generation sequencing or tiled microarray approaches for user defined regions of interest. We validated ArchAlign using well characterized functional elements, and used it to explore the chromatin structural architecture at CTCF binding sites in the human genome. Social media platforms today allow professional footballers to reach millions of people through private marketing as individuals, thereby creating their own brand. The brand and reach of each player is also a valuable resource for clubs in terms of player value and transfer activity. The outlined results in the following article show indicate a positive correlation between the social media value of professional players and the transfer activities of football clubs. Consequently, the impact of digitization on professional football can be shown by a relationship which has not been investigated in research to date

    Using a Full Spectral Raytracer for Calculating Light Microclimate in Functional-Structural Plant Modelling

    Raytracers that allow the spatially explicit calculation of the fate of light beams in a 3D scene allow the consideration of shading, reflected and transmitted light in functional-structural plant models (FSPM). However, the spectrum of visible light also has an effect on cellular and growth processes. This recently created the interest to extend this modelling paradigm allowing the representation of detailed spectra instead of monochromatic or white light and to extend existing FSPM platforms accordingly. In this study a raytracer is presented which supports the full spectrum of light and which can be used to compute spectra from arbitrary light sources and their transformation at the organ level by absorption, reflection and transmission in a virtual canopy.     Density functional theory study of Pd aggregation on a pyridine-terminated self-assembled monolayer

Y.By using density functional theory calculations, the initial steps towards Pd metal cluster formation on a pyridine‐terminated self‐assembled monolayer (SAM) consisting of 3‐(4‐(pyridine‐4‐yl)phenyl)propane‐1‐thiol on an Au(1 1 1) surface are investigated. Theoretical modelling allows the investigation of structural details of the SAM surface and the metal/SAM interface at the atomic level, which is essential for elucidating the nature of Pd–SAM and Pd–Pd interactions at the liquid/solid interface and gaining insight into the mechanism of metal nucleation in the initial stage of electrodeposition. The structural flexibility of SAM molecules was studied first and the most stable conformation was identified, planar molecules in a herringbone packing, as the model for Pd adsorption. Two binding sites are found for Pd atoms on the pyridine end group of the SAM. The strong interaction between Pd atoms and pyridines illustrates the importance of SAM functionalisation in the metal nucleation process. Consistent with an energetic driving force of approximately −0.3 eV per Pd atom towards Pd aggregation suggested by static calculations, a spontaneous Pd dimerisation is observed in ab initio molecular dynamic studies of the system. Nudged elastic band calculations suggest a potential route with a low energy barrier of 0.10 eV for the Pd atom diffusion and then dimerisation on top of the SAM layer.Publisher PDFPeer reviewe
